Elevating Metabolomics Research with the Thermo Scientific™Orbitrap™ Excedion™ Pro

The Thermo Fisher Scientific™ Orbitrap™ Excedion™ Pro represents the next generation of hybrid high-resolution mass spectrometers, introducing features designed to enhance metabolomics research. A notable innovation is a novel scanning mode – enhanced Dynamic Range (eDR) which increases detection capabilities for low-abundance compounds while concurrently mitigating saturation effects from high-abundance species. Data processing is streamlined via Thermo Scientific™ Compound Discoverer™ 3.4 software, which leverages the mzCloud™ 2.0 database to improve compound identification.
The system features a new ion source with increased stability and an electrodynamic ion funnel optimized for labile species. As a result, in performance evaluations, the Orbitrap™ Excedion™ Pro demonstrated a visible reduction in unintentional MS1 fragmentation, leading to higher confidence in compound identification. The integration with Compound Discoverer™ 3.4 further facilitated the automatic detection and grouping of in-source fragments, thereby reducing false positives.
Operationally, the transition to a single dry pump from a traditional two-oil-pump configuration resulted in 30% less energy consumption and reduced noise and heat production.
In the evaluation of an example plant extract dataset, the eDR enabled a more than two-fold increase in detected compounds and a 245% increase in compounds triggered for MS2 fragmentation, which contributed to a 50% increase in mzCloud matches.
Overall, the Thermo Fisher Scientific™ Orbitrap™ Excedion™ Pro delivers a set of improvements designed specifically for metabolomics workflows. These collectively contribute to more accurate and confident metabolite identification and quantitation, making it a valuable tool for researchers in the field.
